中科院营养代谢与食品安全重点实验室,肿瘤代谢及细胞极性的分子调控机制课题组组长

研究方向:极性蛋白(polarity protein)在调控上皮细胞特性以及上皮细胞稳态调控中的作用,以及细胞极性的丢失在肿瘤生成中的机制。运用细胞3维培养系统以及上皮组织类器官模型,探索细胞极性与上皮细胞命运调控,以及上皮细胞极性丢失与上皮细胞恶性转化,及其转移等肿瘤生物学特征之间的关联。基于此研究,希望发现调控上皮细胞修复的新的极性基因和信号通路。此外希望发现肿瘤早期诊断的新标记物或肿瘤治疗的潜在靶点。

1. 深入探讨上皮细胞中的极性基因功能以及在上皮屏障中的重要分子机制,上皮细胞命运调控过程中极性信号的关键机制,比如在自噬以及坏死性凋亡(necroptosis)中的调控机制,并探讨上皮细胞屏障破坏和和修复过程中极性基因的调控机制,以及上皮细胞屏障破坏与肿瘤发生的作用机制研究。
2. 以肿瘤原位移植技术结合生物信息学,分子生物学方法,获得肿瘤在肺部转移和定值中的关键分子节点,上皮细胞EMT和MET转化过程中极性信号的调控机制,以及在远端转移中的相关机制。
3. 以目前先进的类器官技术(organoid),进一步探讨上皮细胞稳态(Homeostasis)打破与损伤修复中极性通路的新功能,来研究肿瘤早期发生中与肿瘤微环境的相互作用,从而明确极性信号在决定上皮细胞命运以及异质性中的分子机制。
